Key Research Findings

  • TRIUMPH-1 (May 2026) — Eli Lilly reported average weight reduction of 28.3% at 80 weeks and 30.3% at 104 weeks across 2,339 participants. All doses met their primary and key secondary endpoints.
  • TRIUMPH-4 (December 2025) — up to 28.7% weight reduction alongside reductions in knee pain in adults with obesity and knee osteoarthritis.
  • TRIUMPH-3 (2026) — reported in adults with obesity or overweight and established cardiovascular disease.
  • Seven further phase 3 readouts are due in 2026; Lilly plans a US regulatory submission in Q1 2027.

Overview

  • A synthetic peptide developed by Eli Lilly, known in research literature as LY-3437943 or “Triple G”.
  • Acts on three hormone receptors at the same time — that combination is what makes it unusual among metabolic peptides.
  • Studied for body weight, blood sugar control, liver fat and heart-related measures.

How It Works

  • Triggers three receptors that normally respond to gut and pancreatic hormones: GLP-1, GIP and glucagon.
  • GLP-1 signal — reduces appetite, slows stomach emptying, and helps insulin release when blood sugar rises.
  • GIP signal — supports the insulin response and changes how fat cells handle energy.
  • Glucagon signal — increases the rate at which the body burns energy and releases stored fat.
  • Binds most strongly to the GIP receptor of the three.
  • A fatty-acid chain attached to the peptide makes it stick to proteins in the blood, so it stays active long enough for once-weekly dosing in trials.

Research Effects

Human Clinical Trials

Body Weight Reduction

Average reductions of 28.3% at 80 weeks and 30.3% at 104 weeks reported in the phase 3 TRIUMPH-1 trial.

Human Clinical Trials

Blood Sugar Control

Improvements in glycaemic measures and A1C reported across phase 2 and phase 3 trials.

Human Clinical Trials

Liver Fat

Reductions in liver fat content reported in clinical research.

Human Clinical Trials

Cholesterol & Triglycerides

Changes in triglycerides and LDL reported in published trial data.

Human Clinical Trials

Appetite & Fullness

Reduced appetite and altered fullness signalling described in trial reports.

Human Clinical Trials

Cardiometabolic Measures

Reported in TRIUMPH-3, in adults with obesity or overweight and established cardiovascular disease.

Lab & Animal Studies

Energy Burn Rate

Increased energy expenditure attributed to glucagon receptor activity, characterised mainly in laboratory and animal studies.

Human Clinical Trials — reported in published human trial dataLab & Animal Studies — laboratory or animal studies only

Amino Acid Sequence

YAQGTFTSDYSILLDKKAQAAFIEYLLEGGPSSGAPPPS-NH239-amino acid synthetic peptide (single-letter code). Modifications: Aib = 2-aminoisobutyric acid at positions 2 and 20; α-Me-Leu = α-methyl-L-leucine at position 13; Lys17 carries a C20 fatty diacid–γ-Glu–AEEA spacer side chain; C-terminal serinamide.
